Solubilized Pancreatic Extracellular Matrix from Juvenile Pigs Protects Isolated Human Islets from Hypoxia-Induced Damage: A Viable Option for Clinical Islet Transplantation

چکیده
The pancreatic extracellular matrix (ECM) is an enormously complex construct. Previous studies underline the challenges to identify optimal combinations and ratios of individual ECM proteins for promoting survival function isolated transplanted islets. This study aimed on assessing efficiency solubilized natural extracted from juvenile pigs, unlimited donor source. Isolated human islets were cultured under a hypoxic atmosphere (2% oxygen) in media supplemented with either porcine (ppECM) or mixture composed collagen-IV, laminin-521, nidogen-1 (hEPM). Control identical conditions without ECM-compounds. Reactive oxygen species production increased three-fold controls but was reduced by hEPM ppECM. Early apoptosis remained preculture levels when treated Preculture viability preserved ppECM administered. Whilst failed respond glucose challenge, treatment physiological insulin response. In summary, overall significantly highest ppECM-treated presents new approach protect hypoxia-induced damage supplementing findings may also serve as starting point novel encapsulation technique

Extracellular vesicles from human pancreatic islets suppress human islet amyloid polypeptide amyloid formation.
Extracellular vesicles (EVs) are small vesicles released by cells to aid cell-cell communication and tissue homeostasis. Human islet amyloid polypeptide (IAPP) is the major component of amyloid deposits found in pancreatic islets of patients with type 2 diabetes (T2D). IAPP is secreted in conjunction with insulin from pancreatic β cells to regulate glucose metabolism. Here, using a combination ...
متن کاملAngiopoietin-1 production in islets improves islet engraftment and protects islets from cytokine-induced apoptosis.
Successful islet transplantation depends on the infusion of sufficiently large quantities of islets, but only a small fraction of implanted islets become engrafted. The underlying mechanisms remain elusive. To probe the mechanism of islet revascularization, we determined the effect of angiopoietin-1 (Ang-1), a proangiogenic and antiapoptotic factor, on the survival, function, and revascularizat...
متن کاملOverexpression of IRS2 in isolated pancreatic islets causes proliferation and protects human beta-cells from hyperglycemia-induced apoptosis.
Studies in vivo indicate that IRS2 plays an important role in maintaining functional beta-cell mass. To investigate if IRS2 autonomously affects beta-cells, we have studied proliferation, apoptosis, and beta-cell function in isolated rat and human islets after overexpression of IRS2 or IRS1. We found that beta-cell proliferation was significantly increased in rat islets overexpressing IRS2 whil...
متن کاملIntroducing a New Experimental Islet Transplantation Model using Biomimetic Hydrogel and a Simple High Yield Islet Isolation Technique
Background: Islet transplantation could be an ideal alternative treatment to insulin therapy for type 1 diabetes Mellitus (T1DM). This clinical and experimental field requires a model that covers problems such as requiring a large number of functional and viable islets, the optimal transplantation site, and the prevention of islet dispersion. Hence, the methods of choice for isolation of functi...
متن کاملLentinan protects pancreatic β cells from STZ‐induced damage
Pancreatic β-cell death or dysfunction mediated by oxidative stress underlies the development and progression of diabetes mellitus (DM). In this study, we evaluated the effect of lentinan (LNT), an active ingredient purified from the bodies of Lentinus edodes, on pancreatic β-cell apoptosis and dysfunction caused by streptozotocin (STZ) and the possible mechanisms implicated. The rat insulinoma...
